Our results indicate that PFA with around 11 fluorine atoms significantly improves aptamer internalization both <i>in vitro</i> and <i>in vivo</i> settings. However, we also observed that the use of PFA tags containing 19 and 23 fluorine atoms on aptamers resulted in nonspecific cell anchoring in control cell lines, affecting the specificity of aptamers. Overall, we found that using a chemical modification strategy could enhance the deep tumor penetration ability of aptamers and validate their effectiveness <i>in vivo</i>.
